Abstract:
The Mitsunobu reaction is a cornerstone of modern post-graduate synthetic planning, offering a highly reliable method for the stereospecific conversion of primary and secondary alcohols into esters, ethers, amines, and thioethers [1, 4]. Driven by the formidable thermodynamic formation of a triphenylphosphine oxide (P=O) bond, this transformation operates under mild, neutral conditions and is distinguished by its strict SN2 mechanism, resulting in the complete inversion of stereochemistry [1, 2, 7]. This atlas outlines the precise mechanistic sequence—from the formation of the Morrison-Brunn-Huisgen betaine to the ultimate displacement of the alkoxyphosphonium intermediate—while mapping out seven structurally diverse applications critical for CSIR NET and GATE advanced curricula [1-4, 8].
